Today in lab we made a broth media using deionized water (DH2O2) and chemical powder. We used a specific math formula, which help determine water to media ratio. . We then heated the broth to dissolve all the powder, evenly distributed the broth among 6 tubes, and in the autoclave to sterilize it to provide the optimal environment for our bacteria to grow in the future.

Methods

* Turn on hot plate

* Calculate amount of powder needed using the formula below

Total DH2O2 X Grams of media needed = Media per tube = amount of media need

needed Per 1 Liter DH2O2 1 liter DH2O2

(1000ml)

DH2O2= 48 ml- for all 6 tubes (8ml per tube)

Media = 23 grams per liter

48 X 23g =1.104g (or 1.1g)

* Weigh Boat (bowl to weigh powder in)before adding powder . Boat = 1.9g

* Place scale to 3g (1.9g boat + 1.1g powder = 3g). Add powder using metal spatula until scale is balanced

* Add 45ml of DH2O2 into a flask, using 5 ml pipette (9 times)

* Pour powder into flask.

* With the remaining 3 ml of DH2O2, using the 1ml pipette, put 1 ml into boat at a time making sure to get all powder off the boat.

* Pour mixture (powder and 3ml DH2O2) into flask with the other media mixture.

* Stir broth with a glass stirrer, making sure to scrap the bottom of the flask

* Place flask on hot plate.

* Let boil/cook for a few minute until powder is completely dissolved.

* Keep a close eye so broth does not boil over.

* Remove flask from hot plate with gloves.

* Using the 5ml and 1ml pipette dispense 8ml of broth into each of the 6 tubes.

* Label each tube with autoclave tape ad write name, date and substance in tubes.

* Place the 6 tubes into a rack and then into the autoclave machine.
